Calibration involves temperature and CO₂ concentration. For temperature: place a high-precision thermometer (±0.1°C) inside, let it stabilize, compare readings, and adjust the setpoint if needed (tolerance: ±0.2°C). For CO₂: connect a calibrated CO₂ analyzer to the sampling port, compare its reading to the display, and adjust (tolerance: ±0.1% CO₂). Always let the incubator stabilize for 30 min before calibration. Refer to the manufacturer's manual for specific procedures.
First, turn off power and CO₂ supply, and remove all cultures, shelves, and water pan. Wipe all interior surfaces, door gasket, and shelves with 70% ethanol or mild soap solution. Rinse with sterile distilled water to remove residue. Clean the water pan weekly — empty, wipe with 70% ethanol, and refill with sterile distilled water. For deep decontamination, use a high-temperature sterilization cycle (if available) or wipe with 3‰ benzalkonium chloride, then rinse thoroughly.
CO₂ incubators are primarily used for cell and tissue culture in biomedical research. They maintain a controlled environment at 37°C, 5% CO₂, and ~95% humidity, creating optimal pH (7.2–7.4) for mammalian cell growth. Common applications include cancer research, IVF, stem cell engineering, vaccine production, drug screening, and hybridoma antibody production.
